- Several voice actors from the previous Inuyasha series reprise their roles in the dub of Yashahime including:
- Richard Ian Cox as Inuyasha
- Kira Tozer as Kagome Higurashi

- Kirby Morrow as Miroku
- Kirby, however, only voiced Miroku in the first episode before his passing on November 18, 2020, 2 weeks later.
- Jillian Michaels as Shippo
- Paul Dobson as Myoga
- David Kaye as Sesshomaru, who returns to voice him again after he didn't return in The Final Act.
- Linda Darlow as Kaede
- Brenna O'Brien as Rin
- Willow Johnson as Kikyo
- Cathy Weseluck as Mrs. Higurashi
- Terry Klassen as Hachiemon
- and Richard Newman as Tōtōsai
- However, some actors from the dubs of Inuyasha and Inyuasha: The Final Act did not reprise their roles for Yashahime and were replaced by new voice actors:
- Alan Lee replaced Aidan Drummond as Kohaku.
- Andrew Russell replaced Don Brown as Jaken.
- William Frederick Knight replaced French Tickner as Grandpa Higurashi.
- Ian James Corlett replaced Kirby Morrow as Miroku, following the latter's passing on November 18.
- Janice Kawaye replaced Nicole Bouma as Shiori.

- Mick Lauer replaced Don Brown as Tōga.
- This is the first anime dub David Kaye appeared in since Ghost in the Shell: Stand Alone Complex 2nd GIG: Individual Eleven, it is also David's first Los Angeles anime dub.
- This was Kirby Morrow's final voice acting role as he passed away on November 18 which was almost two weeks after the English dub was released on FUNimationNow. He was replaced by Ian James Corlett in his first anime role since Mobile Suit Gundam Wing.
